Research focus

Prof. Pasqualucci’s research focuses on the genetic and epigenetic basis of B-cell malignancies, with particular emphasis on the two most common subtypes: diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma (FL). Her research centers on identifying and functionally characterizing the genetic alterations associated with these tumors, including through their in vivo modeling, with the ultimate goal of discovering novel biomarkers and laying the groundwork for more effective therapeutic strategies.

Prof. Pasqualucci’s laboratory integrates genomic analyses, multi-omics approaches, functional genomics, and genetically engineered mouse models to elucidate the mechanisms driving the malignant transformation of germinal center (GC) B cells. A major area of current research is understanding the epigenetic dysfunction caused by genetic lesions in FL and DLBCL. These include mutations in histone and chromatin modifier genes and aberrant somatic hypermutation of super-enhancer regions, which her group identified as recurrent mutational targets in both FL and DLBCL.

Prof. Pasqualucci’s studies established the acetyltransferase CREBBP and the methyltransferase KMT2D as bona fide tumor suppressors in GC-derived lymphomas, with their disruption occurring early in tumor clonal evolution. This knowledge is currently being leveraged to develop targeted therapeutic approaches for these diseases.

The Lymphomagenesis Laboratory is supported by a five-year startup grant from the Horten Health Foundation.
